INTENDED USE
Avian Leukosis Antigen Rapid Test Cassette (Meconium/Cloaca Swab/Egg White) is a sandwich lateral flow immunochromatographic assay for the qualitative detection of Avian Leukosis Antigen (ALV Ag) in chicken’s meconium, cloaca
swab or egg white sample.
PRINCIPLE
Avian Leukosis Antigen Rapid Test Cassette is based on sandwich lateral flow immunochromatographic assay. The test cassette has a testing window with an invisible T (test) zone and C (control) zone. When sample is applied into the
sample well on the cassette, the liquid will laterally flow on the surface of the test strip. If there is enough Avian Leukosis Antigen in the sample, a visible T line will appear. The C line should always appear after a sample is applied, indicating a
valid result. By this means, the test can accurately indicate the presence of ALV Ag in the sample.
STORAGE AND STABILITY
Store as packaged in the sealed pouch either at room temperature or refrigerated (2-30 °C). The test is stable through the expiration date printed on the sealed pouch. The test cassette must remain in the sealed pouch until use. DO NOT FREEZE. Do not use beyond the expiration date.
PRECAUTIONS
Do not use after the expiration date. Do not reuse the test kit.
Handle all specimens as if they contain infectious agents. Observe established precautions against microbiological hazards throughout testing and follow the standard procedures for proper disposal of specimens.
Wear disposable gloves and eye protection when specimens are being tested.
Humidity and temperature can adversely affect results.
Do not remove test cassette from its pouch until immediately before use.
Do not mix components from different lot and different products.

DIRECTIONS FOR USE
Allow the test cassette, specimen, buffer and/or controls to equilibrate to room temperature (15-30 °C) prior to testing.
1. Sample processing
1.1 Meconium/Cloaca swab
Collect meconium/cloaca sample with the swab provided. Insert the wet swab into the provided plastic tube with diluent buffer. Rotate the swab for approximately 10 seconds while pressing the head against the inside of the tube to release the antigen in the swab. Remove the swab while squeezing the swab head against the inside of the tube as you remove it to expel as much liquid as possible from the swab. Leave the tube for 1 minute, and then take the supernatant (diluent sample) for testing.
1.2 Egg White
Take a whole egg, crack the shell at the air chamber to extract the egg white.
Use a pipette: Pipette 50 μL of egg white and 150 μL of diluent buffer into the tube; mix the specimen and the buffer well.
Use a dropper: Hold the dropper vertically and transfer 2 drops of egg white (approximately 50 μL) and 8 drops of diluent buffer (approximately 150 μL) into the tube; mix the specimen and the buffer well.
Note: Use a clean dropper to absorb the buffer every time.
2. Place the test cassette on a clean, flat surface.
3. Hold the dropper vertically and transfer 4 drops of diluent sample (approximately 100μL) to the specimen well (S) of the test cassette, and then start the timer.
Note: If the liquid runs too slowly, it is recommended to add another 2-3 drops of diluent sample.
4. Read the result at 10 minutes. Do not interpret results after 15 minutes.
